Electrochemical conversion of biomass-based oxygenated compounds

Dwindling fossil fuel resources and substantial release of CO2 from their processing have increased the appeal to use biomass as a sustainable platform for synthesis of chemicals and fuels. Steps toward this will require selective upgrading of biomass to suitable intermediates. Traditionally, biomass upgrading has involved thermochemical processes that require excessive amounts of petrochemical-derived H2 and suffer from poor product selectivity. Electrochemical routes have emerged as promising alternatives because of (a) the replacement of petrochemical-derived H2 by protons generated in situ, (b) mild operating temperatures and pressures, and (c) the use of electrode potential to tune reaction rates and product selectivity. In this review, we highlight the advances in the electrocatalytic hydrogenation and oxidation of biomass-derived platform molecules. The effects of important reaction parameters on electrochemical efficiency and catalytic activity/selectivity are thoroughly discussed. We conclude by summarizing current challenges and discussing future research directions.

Discovery and optimization of novel N-benzyl-3,6-dimethylbenzo[d]isoxazol-5-amine derivatives as potent and selective TRIM24 bromodomain inhibitors with potential anti-cancer activities

Tripartite motif-containing protein 24 (TRIM24), recognized as an epigenetic reader for acetylated H3K23 (H3K23ac) via its bromodomain, has been closely involved in tumorigenesis or tumor progression of several cancers. Developing inhibitors of TRIM24 is significant for functional studies and drug discovery. Herein, we report the identification, optimization and evaluation of N-benzyl-3,6-dimethylbenzo[d]isoxazol-5-amines as TRIM24 bromodomain inhibitors starting from an in house library screening. Structure-based optimization leads to two potent and selective compounds 11d and 11h in an Alphascreen assay with IC50 values of 1.88 muM and 2.53 muM, respectively. The viability assay demonstrates the great potential of this series of compounds as inhibitors of proliferation of prostate cancer (PC) cells LNCaP, C4-2B. A colony formation assay further supports this inhibitory activity. Compounds 11d and 11h inhibit cell proliferation of other cancer types such as non-small cell lung cancer (NSCLC) cells A549 with IC50 values of 1.08 muM and 0.75 muM, respectively. These data suggests that compounds 11d and 11h are promising lead compounds for further research.

Metabolomic analysis of blood plasma after oral administration of N-acetyl-D-glucosamine in dogs

N-acetyl-D-glucosamine (GlcNAc) is a monosaccharide that polymerizes linearly through (1,4)-beta-linkages. GlcNAc is the monomeric unit of the polymer chitin. GlcNAc is a basic component of hyaluronic acid and keratin sulfate found on the cell surface. The aim of this study was to examine amino acid metabolism after oral GlcNAc administration in dogs. Results showed that plasma levels of ectoine were significantly higher after oral administration of GlcNAc than prior to administration (p < 0.001). To our knowledge, there have been no reports of increased ectoine concentrations in the plasma. The mechanism by which GlcNAc administration leads to increased ectoine plasma concentration remains unclear; future studies are required to clarify this mechanism. Do you like my blog? Selective hydrogenolysis of 2-furancarboxylic acid to 5-hydroxyvaleric acid derivatives over supported platinum catalysts

The conversion of 2-furancarboxylic acid (FCA), which is produced by oxidation of furfural, to 5-hydroxyvaleric acid (5-HVA) and its ester/lactone derivatives with H2 was investigated. Monometallic Pt catalysts were effective, and other noble metals were not effective due to the formation of ring-hydrogenation products. Supports and solvents had a small effect on the performance; however, Pt/Al2O3 was the best catalyst and short chain alcohols such as methanol were better solvents. The optimum reaction temperature was about 373 K, and at higher temperature the catalyst was drastically deactivated by deposition of organic materials on the catalyst. The highest yield of target products (5-HVA, delta-valerolactone (DVL), and methyl 5-hydroxyvalerate) was 62%, mainly obtained as methyl 5-hydroxyvalerate (55% yield). The byproducts were mainly ring-hydrogenation compounds (tetrahydrofuran-2-carboxylic acid and its ester) and undetected ones (loss of carbon balance). The catalyst was gradually deactivated during reuses even at a reaction temperature of 373 K; however, the catalytic activity was recovered by calcination at 573 K. The reactions of various related substrates were carried out, and it was found that the O-C bond in the O-CC structure (1,2,3-position of the furan ring) is dissociated before CC hydrogenation while the presence and position of the carboxyl group (or methoxy carbonyl group) much affect the reactivity.

Cytotoxic and Apoptosis-inducing Activities of Taraxastane-type Triterpenoid Derivatives in Human Cancer Cell Lines

Twenty-eight taraxastane-type triterpenoid derivatives 4???31 were prepared from the naturally occurring triterpenoids faradiol (1) and heliantriol C (3). The cytotoxic activities of these compounds and arnidiol (2) were evaluated in leukemia (HL60), lung (A549), duodenal (AZ521), and breast (SK-BR-3) cancer cell lines. 21-Oxoarnidiol (18) and faradiol 3,16-di-O-l-alaninate (31) exhibited potent cytotoxicity, with 50% inhibitory concentrations of 0.5???2.7?mum. In particular, flow cytometric analysis indicated that compound 31 induced typical apoptotic cell death in HL60 cells. These results suggested that taraxastane-type triterpenoid derivatives might provide useful antitumor agents with apoptosis-inducing activity.
