Indirubins have been identified as potent ATP-competitive protein kinase inhibitors. Structural modifications in the 5-and 3?-position have been extensively investigated, but the impact of substituents in 5?-position is not equally well-studied. Here, we report the synthesis of new indirubin 3?-and 5?-derivatives in the search of water-soluble indirubins by introducing basic centers. Antiproliferative activity of all compounds in tumor cells was evaluated along with kinase inhibition of selected compounds. The results show the 3?-position to tolerate large substituents without compromising activity, whereas bulk and rigid substituents in 5?-position appear unfavorable. Screening molecular targets of water-soluble 3?-oxime ethers revealed 6ha as preferential inhibitor of insulin-like growth factor 1 receptor (IGF-1R) in a panel of 22 protein kinases and in cells. Consistently, 6ha inhibited tumor cell growth in the NCI 60 cell line panel and induced apoptosis. The results indicate that the 5?-position provides limited space for chemical modifications and identify 6ha as a potent water-soluble indirubin-based IGF-1R inhibitor.

The aim of this study was the synthesis and lead structure selection of a best anti-leukemic agent from a library of aza-podophyllotoxin analogues (APTs). To this end, we report a scalable, modified multicomponent reaction using a “sacrificial” aniline partner as a more general route to rapidly construct the pivotal library of 50 APT analogues. Our preliminary structure activity relationship studies for anti-leukemic activity also address the innate toxicity of these compounds against non-malignant cells. As a result, we identified 2 novel compounds 2ca? and 2jc? more potent than etoposide 1 (25-60 fold) having high selectivity against the human THP-1 leukemia cell line and a minimal toxicity (IC50 of 9.3 ± 0.8 and 19.6 ± 1.4 nM respectively) which represent the best candidates for further pharmacological optimization.

It is shown how secondary alcohols are oxidized to provide the corresponding ketones by use of Oxone and solid-supported hypervalent iodine catalysts. Under experimentally simple conditions with acetonitrile at elevated temperatures, excellent conversions were achieved with low catalyst loadings (0.2-5 mol%) when employing the conjugates 5 and 6 derived from IBX and IBS. The catalysts are broadly applicable to a range of alcohol substrates. Of primary importance with respect to sustainability issues, the metal-free catalysts are easily removed from the reaction mixture through filtration, and they can be re-used in oxidation processes for multiple times, without loss of catalytic activity.

This is based on immersion of a solid-phase microextraction fiber of PDMS/DVB into the oil matrix, followed by Gas Chromatography/Mass Spectrometry. Furthermore, for the first time, certain fatty amides, gamma- and delta-lactones of high molecular weight, and other aromatic compounds such as some esters derived from cinnamic acid have been detected in edible oils.
