Biocatalytic Enantioselective Hydroaminations for Production of N-Cycloalkyl-Substituted L-Aspartic Acids Using Two C?N Lyases

N-cycloalkyl-substituted amino acids have wide-ranging applications in pharma- and nutraceutical fields. Here we report the asymmetric synthesis of various N-cycloalkyl-substituted l-aspartic acids using ethylenediamine-N,N?-disuccinic acid lyase (EDDS lyase) and a previously engineered variant of methylaspartate ammonia lyase (MAL-Q73A) as biocatalysts. Particularly, EDDS lyase shows broad non-natural substrate promiscuity and excellent enantioselectivity, allowing the selective addition of homo- and heterocycloalkyl amines (comprising four-, five- and six-membered rings) to fumarate, giving the corresponding N-cycloalkyl-substituted l-aspartic acids with >99% e.e. This biocatalytic methodology offers an alternative synthetic choice to prepare difficult N-cycloalkyl-substituted amino acids. Given its very broad amine scope, EDDS lyase is an exceptionally powerful synthetic tool that nicely complements the rapidly expanding toolbox of biocatalysts for asymmetric synthesis of noncanonical amino acids. Synthesis and fungicidal activities of perfluoropropan-2-yl-based novel quinoline derivatives

A series of novel perfluoropropan-2-yl-based quinoline derivatives was designed and synthesized utilizing tebufloquin as the lead compound. The structures of all the newly synthesized compounds were confirmed by spectroscopic data 1HNMR, MS and elemental analysis. The results of bioassay indicated that these compounds exhibited potent fungicidal activities against Erysiphe graminis. Especially, compound 8c displayed excellent activity with EC50 value at 1.48 mg / L, which was better than that of the commercialized fungicide-tebufloquin. The structure-activity relationship for these new compounds was also discussed.

The synthesized polymers Poly [(1,3-dioxane-5-oxy) (1,3-dioxolane-4-methoxy)phosphazene] (DOPP) and Poly[bis(2-Tetrahydro-3-furanoxy)phosphazene] (THFPP) were mixed with varied amounts of lithium bis(trifluoromethane)sulfonamide (LiTFSI) and the interactions between the salt and the polymer chains were studied by Fourier transform infrared (FT-IR) and differential scanning calorimetry (DSC) measurements. Electrochemical characterization was performed by electrochemical impedance spectroscopy (EIS) and direct current polarization in the temperature range of 20?60 C. These measurements were utilized to calculate the lithium transference number (t+), the lithium conductivity (sigma) and its activation energy in order to elucidate the lithium transport behavior. Relatively high lithium transference numbers of 0.6 (DOPP) and 0.7 (THFPP) at 60 C are found and reveal maximum lithium conductivities of 2.8·10?6 S?cm?1 and 9.0·10?7 S?cm?1 for DOPP and THFPP at 60 C, respectively.

Structural identification of binary tetrahydrofuran + O2 and 3-hydroxytetrahydrofuran + O2 clathrate hydrates by rietveld analysis with direct space method

The structural determination of clathrate hydrates, nonstoichiometric crystalline host-guest materials, is challenging because of the dynamical disorder and partial cage occupancies of the guest molecules. The application of direct space methods with Rietveld analysis can determine the powder X-ray diffraction (PXRD) patterns of clathrates. Here, we conducted Rietveld analysis with the direct space method for the structural determination of binary tetrahydrofuran (THF) + O2 and 3-hydroxytetrahydrofuran (3-OH THF) + O2 clathrate hydrates in order to identify the hydroxyl substituent effect on interactions between the host framework and the cyclic ether guest molecules. The refined PXRD results reveal that the hydroxyl groups are hydrogen-bonded to host hexagonal rings of water molecules in the 512 64 cage, while any evidences of hydrogen bonding between THF guests and the host framework were not observed from PXRD at 100 K. This guest-host hydrogen bonding is thought to induce slightly larger 512 cages in the 3-OH THF hydrate than those in the THF hydrate. Consequently, the disorder dynamics of the secondary guest molecules also can be affected by the hydrogen bonding of larger guest molecules. The structural information of binary clathrate hydrates reported here can improve the understanding of the host-guest interactions occurring in clathrate hydrates and the specialized methodologies for crystal structure determination of clathrate hydrates.

Design and synthesis of novel, potent and selective hypoxanthine analogs as adenosine A1receptor antagonists and their biological evaluation

Multipronged approach was used to synthesize a library of diverse C-8 cyclopentyl hypoxanthine analogs from a common intermediate III.
