As a kind of biomass resource, lignin is the only natural renewable resource that can provide abundant aromatic compounds, which is undoubtedly considered as the best candidate to produce value-added chems. by lignin depolymerization Thus, it is of great significance to develop approaches that are able to accommodate the complex structure of lignin. Herein, an efficient depolymerization method is implemented in methanol using a mesoporous solid superbase KF/γ-Al2O3 catalyst. Research shows that the depolymerization of lignin is not obvious below the critical temperature of methanol, and the conversion of lignin increases significantly above the critical temperature, which can reach 60.6% at 270°C. Phenols and methoxy-substituted monocyclic aromatic compounds are predominant in the liquid products acquired by gas chromatog./mass spectrometry. Furthermore, oligomers were analyzed by time-of-flight mass spectrometry equipped with electrospray ionization. It is found that the mol. mass of the oligomers varied between 200 and 600 Da, the degree of unsaturation is mainly 4 and 8 with 1-2 aromatic rings, and the carbon number is found to be within the range of 15-30. It is speculated that the oligomers are mainly monophenyl or biphenyl compounds containing branched chains.

Somatic cells can be reprogrammed into pluripotent stem cells (PSCs) by using pure chems., providing a different paradigm to study somatic reprogramming. Moreover, precise manipulation of the cell fate transition in a step-wise manner through the XEN-like state allowed the authors to identify small-mol. boosters and establish a robust chem. reprogramming system with a yield up to 1000-fold greater than that of the previously reported protocol. The total phenolic content was determined and the antioxidant activity of the WV was measured. The total phenolic contents of PSWV and HSDSWV were 75.9 and 98.1 mg/L, resp., when they were expressed as catechol equivalent In 1,1-diphenyl-2-picrylhydrazyl assay, free radical scavenging activity of HSDSWV was also superior than that of PSWV. In addition, the antioxidant activities of the two kind of WVs exhi-bited a good pos. correlation with total phenolic content, and the linear correlation coefficients ranged from 0.9594 to 0.9970. The two kind of WVs could be used in the fields of food and medicine due to their powerful anti-oxidant activities.

Wood vinegar(WV) has a powerful antioxidant activity, but it is unclear which components are responsi-ble for the antioxidant activity. In the present study, the double-column retention index qual. method was used for the identification of the major components in five kinds of WV. And the major antioxidants of wood vinegar were accurately identified with the aid of Pearson product-moment correlation coefficients and authentic standard samples. Our results demonstrate that phenolic compounds are mainly responsible for the powerful antioxidant activity. 2,6-Dimethoxyphenol is the most powerful antioxidant in WV. 2-Methoxyphenol and 3-methyl-1,2-cyclopentane-dione also have an important influence on the antioxidant activity of WV. Our results suggest that the contents of 2-methoxyphenol, 2,6-dimethoxyphenol and 3-methyl-1,2-cyclopentanedione should act as the criteria for evaluating the antioxidant activity of WV. Our work will provide useful information for WV’s application in the fields of food and medicine as antioxidants.

Glucose and fructose are widely available and renewable resources. They were used to prepare acetic acid (AA) under the catalysis of potassium acetate (KAc) by thermogravimetric anal. and pyrolysis coupled with gas chromatog. and mass spectrometry. The TGA result showed that the KAc addition lowered the glucose’s thermal decomposition temperatures (about 30°C for initial decomposition temperature and 40°C for maximum mass loss rate temperature), implying its promotion of glucose’s decomposition The Py-GC/MS tests illustrated that the KAc addition significantly altered the composition and distribution of hexose pyrolysis products. The maximum yield of AA was 52.1% for the in situ catalytic pyrolysis of glucose/KAc (1:0.25 wt/wt) mixtures at 350°C for 30 s. Under the same conditions, the AA yield obtained from fructose was 48% and it increased with the increasing amount of KAc. When the ratio reached to 1:1, the yield was 53.6%. In comparison, a study of in situ and online catalytic methods showed that KAc can not only catalyze the primary cracking of glucose, but also catalyze the cracking of a secondary pyrolysis stream. KAc plays roles in both phys. heat transfer and chem. catalysis.

Incomplete epigenetic reprogramming of the genome of donor cells causes poor early and full-term developmental efficiency of somatic cell nuclear transfer (SCNT) embryos. Previous research indicate that inhibition of the histone H3 K79 methyltransferase DOT1L, using a selective pharmacol. inhibitor EPZ004777 (EPZ), significantly improved reprogramming efficiency during the generation of mouse induced pluripotent stem cells. However, the roles of DOT1L in porcine nuclear transfer-mediated cellular reprogramming are not yet known. Here we showed that DOT1L inhibition via 0.5 nM EPZ treatment for 12 or 24 h significantly enhanced the blastocyst rate of SCNT embryos and dramatically reduced the level of H3K79me2 during SCNT 1-cell embryonic development. Addnl., H3K79me2 level in the EPZ-treated SCNT embryos was similar to that in in vitro fertilized embryos, suggesting that DOT1L-mediated H3K79me2 is a reprogramming barrier to early development of porcine SCNT embryos. qRT-PCR anal. further demonstrated that DOT1L inactivation did not change the expression levels of DOT1L itself but increased the expression levels of POU5F1, LIN28, SOX2, CDX2 and GATA4 associated with pluripotency and early cell differentiation. In conclusion, DOT1L inhibitor improved early developmental efficiency of porcine SCNT embryos probably via inducing the increased expression of genes important for pluripotency and lineage specification.

Digital polymers with precise arrangements of units acting as the 0- or 1-bit, have shown bright promise for the data storage media. The easiness of encoding and decoding these polymers is essential when putting them into practical applications. Herein, the concept of utilizing sequence building block and sequencing indicator to generate easily encodable and decodable digital polymers is proposed. With the thiol-maleimide Michael coupling for digital chain growth, the dithiosuccinimide (DTS) motifs generated from the succinimide thioether linkages are used as the sequence building block. The DTS can be equipped with different groups, thus greatly enabling the modular encoding process. Importantly, the tandem mass spectrometry (MS/MS) results unveil the relatively abundant DTS-induced MS signal, which facilitates the decoding process with DTS acting as the sequencing indicator. The synergetic MS/MS induced cleavages of the two C-S bonds on DTS are responsible for the relatively abundant MS signal. Finally, the work exemplifies the quant. decoding of a digital polymer with DTS as the sequence building block and sequencing indicator. This study demonstrates the concept of sequence building block and sequencing indicator, which advances the digital polymers for practical applications.

Swine and chicken manures are abundant solid wastes that can be converted into carbonaceous materials through hydrothermal carbonization (HTC). Owing to their unique biochem. compositions, co-HTC of these two types of manures may have significant implications for the generated products. We investigated the co-HTC of swine manure and chicken manure to understand the influence of the interaction between contrasting manures on the properties of the derived products. The results indicated that co-HTC treatment enhanced the formation of solid product and improved the C and N contents, heating value, and energy yield of the resulting hydrochar. Regarding the ignition temperature and comprehensive combustion index, the combustion properties of the hydrochar were enhanced owing to the mutual effect of the HTC intermediates. Addnl., the interaction of the intermediates significantly impacted the transfer of nitrogenous species and generation of organic acids and organic polymers with fused-ring structures. Therefore, co-HTC processing of animal manures could potentially provide a sustainable pathway for the conversion of animal waste into solid products with improved characteristics compared to those produced by treating the two feedstocks sep.
