Impact of freeze-drying and subsequent storage on milk metabolites based on 1H NMR and UHPLC-QToF/MS

This study is the first to investigate the effect of freeze-drying on raw milk metabolites using untargeted metabolomics. To detect as many metabolites as possible in the liquid fractions, a multiplatform metabolomics approach was implemented using both NMR and MS-based techniques. The chemical fingerprint of freeze-dried milk powder was monitored during storage at three temperatures (room temperature, 4 C, ?20 C) for up to 224 days. This study demonstrated that freeze-drying was an efficient means of drying milk resulting in only minor changes to the metabolites. With respect to storage, freeze-dried milk powders stored at 4 C and ?20 C exhibited a stable metabolome while samples stored at 20 C resulted in a clear change. During ambient storage, the concentrations of orotic acid, riboflavin and acetyl-carbohydrate reduced while those of fatty acids, threonic acid and uridine increased. The results from this study provide the foundation for experiment design and marker selection for further studies on milk metabolomics.

Recent Advances: Time-resolved kinetic studies and product analysis have expanded our mechanistic understanding of radical reaction pathways of sulfur-containing amino acids. These reactions are now studied in some detail for Met and Cys in proteins, and homocysteine (Hcy) chemically linked to proteins, and the role of protein radical reactions in physiological processes is evolving. Critical Issues: Radical-derived products from Cys, Hcy, and Met can react with additional amino acids in proteins, leading to secondary protein modifications, which are potentially remote from initial points of radical attack. These products may contain intra- and intermolecular cross-links, which may lead to protein aggregation. Protein sequence and conformation will have a significant impact on the formation of such products, and a thorough understanding of reaction mechanisms and specifically how protein structure influences reaction pathways will be critical for identification and characterization of novel reaction products. Future Directions: Future studies must evaluate the biological significance of novel reaction products that are derived from radical reactions of sulfur-containing amino acids.

Hydrotreating of wheat straw in toluene and ethanol

In the present work, wheat straw was hydroliquefied at a temperature of 300C for 4h in ethanol or toluene in order to obtain bio-components which are useful for fuel purposes. The experiments were performed in a 100mL batch reactor under hydrogen pressure of 70bar. Typically, 2g of straw and 0.1g of catalyst (66%Ni/SiO2-Al2O3) were dispersed in 15g of solvent.
