Identification of aroma compounds in chinese “moutai” and “langjiu” liquors by normal phase liquid chromatography fractionation followed by gas chromatography/olfactometry

The aroma-active compounds in two famous Chinese soy sauce aroma type liquors, Moutai and Langjiu liquors, were investigated in this study. The aroma compounds were isolated using liquid/liquid extraction, and further fractionated into acidic, basic, and neutral fractions. The neural fraction was applied to a normal phase liquid chromatography column and further separated based on their polarity. The aroma compounds of seven fractionations were separately analyzed by GC/Olfactometry (GC/O). A total of 186 aroma-active compounds were identified by GC/O and GC/MS. Among these compounds, ethyl hexanoate, hexanoic acid, 3-methylbutanoic acid, 3-methylbutanol, 2,3,5,6-tetramethylpyrazine, ethyl 2- phenylacetate, 2-phenylethyl acetate, ethyl 3-phenylpropanoate, 4-methylguaiacol, and gamma-decalactone had the highest aroma intensity. Several other basic compounds, including 2,3-dimethyl-5-ethylpyrazine, 2,3,5-trimethyl-6-ethylpyrazine, 2,3,5-trimethylpyrazine, were identified to have high aroma indentify. In addition, vanillin, gamma-heptalactone, gamma-nonalactone, Z-whisky lactone, furaneol, and sotolon, were identified in the liquors. Geosmin was also detected in these two liquors.

Effect of drying and storage on the volatile compounds of jujube fruit detected by electronic nose and GC-MS

Dried jujube fruit has been wildly used as flavoring in food preparation and food industry because of its unique and exquisite flavor. However, there is limited information available on the formation and alteration of jujube flavor during drying and storage. In this study, the effects of drying and storage on the volatiles in jujube fruit were investigated by electronic nose and GC-MS. Electronic nose and GC-MS could distinguish different jujube samples clearly; the ester and aldehydes showed significant decreases during drying, while furans and pyrazines were generated and increased with heat time; the esters, aldehydes, furans and pyrazines showed significant decreases, and some of them even disappeared during storage, while acids and terpenes increased with the extension of storage. Based on the present results, furans and pyrazines, generated during drying, are the predominant components of the unique and exquisite flavor of dried jujube, and these compounds could be negatively in?uenced because of evaporation, oxidation and degradation of jujube volatiles during storage.

Synthesis and biological activity of alpha-methylene-gamma-lactones as new aroma chemicals

Seven kinds of alpha-methylene-gamma-lactones with an alkyl group at the C-4 position were synthesized according to a previously described method, with yields of 28-34%. These alpha-methylene-gamma-lactones had characteristic and unique odors. All alpha-methylene-gamma-lactones added a roast-like odor to materials. The antimicrobial effects of alpha-methylene-gamma-lactones were investigated by using a paper disk diffusion method. The results showed the alpha-methylene-gamma-lactones inhibited the growth of three bacteria (Staphylococcus aureus, Escherichia coli, and Pseudomonas fluorescens) and two fungi (Saccharomyces cerevisiae and Aspergillus niger). In particular, alpha-methylene-gamma-undecalactone and alpha-methylenegamma-dodecalactone exhibited potent inhibition of the growth of these microorganisms compared to butyl p-hydroxybenzoate as standard antibiotic. The umu test revealed that the alpha-methylene-gamma-lactones suppressed the SOS-inducing activity of three mutagens, furylfuramide, UV irradiation, and TrpP-1, respectively. The antimicrobial effects and the suppressive effects of SOS induction by alpha-methylene-gamma-lactones had a tendency to intensify as the number of carbons in the side chain increased.

Direct hydrogenolysis of lactone to carboxylic acid (i.e., hydrogenolysis of the Calkoxy-O bond with the carbonyl group untouched) is generally difficult, as the current strategies employing Br°nsted acids as the catalyst usually require harsh conditions such as a high temperature and a high H2 pressure. Herein, we report a developed solvent-free catalytic transformation, in which W(OTf)6 is believed to promote the hydrogenolysis process. This strategy could efficiently hydrogenate lactones to carboxylic acids under extra mild conditions (e.g., a reaction temperature of <150 C and 1 atm of H2) and showed a broad substrate scope. In addition, the catalytic protocol can be further applied to the hydrogenolysis of polyhydroxyalkanoate, as a renewable polymer, to the corresponding straight-chain carboxylic acids. An extensive mechanistic study was subsequently performed, and the density functional theory calculations revealed a reaction pattern, including the complete cleavage of the C=O bond with the assistance of the W(OTf)6 catalyst. Moreover, the key intermediate created in the mechanism, as an oxonium with an OTf moiety, was successfully detected by electrospray ionization mass spectra. Through a comparison with the Br°nsted acid-catalyzed system, the study confirmed that the existence of the OTf moiety can significantly lower the barriers associated with the rearrangement and elimination processes. Meanwhile, emphasis was placed on the critical role that the anion plays, as well as the fact that the anion effect is directly related to the chemoselectivity. The study explores the complex volatile fraction of extra-virgin olive oil by combining high concentration-capacity headspace approaches with comprehensive two-dimensional gas chromatography, which is coupled with time of flight mass spectrometry. The coverage of both targeted and untargeted 2D-peak-region features, which corresponds to detectable analytes, was examined, while concentration factors (CF) for a selection of informative analytes, including key-odorants and off-odors, and homolog-series relative ratios were calculated and the information capacity was discussed. The results highlighted the differences in concentration capacities, which were mainly caused by polymer-accumulation characteristics (sorptive/adsorptive materials) and its amount. The relative concentration capacity for homologues and potent odorants was also discussed, while headspace linearity and the relative distribution of analytes, as a function of different sampling amounts, was examined. This last point is of particular interest in quantitative studies where accurate data is needed to derive consistent conclusions.

Effect of End-point Temperature and Degree of Doneness on Sensory and Instrumental Flavor Profile of Beefsteaks

Beef M. longissimus thoracis et lumborum was used to investigate the effects of 6 end-point temperatures on flavor and texture. Steaks cooked to medium temperature were associated with 4-heptanolide, whereas those cooked to higher temperatures were associated with pyrazines and hexanol. These steaks were also associated with flavor attributes, such as roasted, burnt, and beefy. However, consumers did not find differences in flavor liking of steaks (P > 0.05) cooked to 6 end-point temperatures. Although steaks cooked to lower temperatures were liked for their tenderness and juiciness, overall liking scores were not affected by end-point temperature. Thus both flavor and tenderness play an important role in consumer satisfaction.
